Product Overview
2,4-Dihydro-4-[4-[4-(4-hydroxyphenyl)-1-piperazinyl]phenyl]-2-(1-methylpropyl)-3H-1,2,4-triazol-3-one is supplied under CAS 106461-41-0, with the molecular formula C₂₂H₂₇N₅O₂ and a molecular weight of approximately 393.49.
It is the whole right-hand side of itraconazole, assembled and waiting.
A triazol-3-one ring carrying a sec-butyl group at N-2 and an aryl group at N-4; that aryl connected through a piperazine to a second benzene ring; and on that second ring, a free phenol.
The phenol is the point. It is the nucleophile that attacks the activated dioxolane fragment, displacing its mesylate and forming the ether bond that joins the two halves of the drug.
Everything else in this molecule is already in its final form and passes into itraconazole unchanged.
Grades and specifications are available on request. Every consignment carries a batch Certificate of Analysis and a GHS-format Safety Data Sheet.

Application in Synthesis
Where It Goes
The phenol is deprotonated and displaces the leaving group on the activated dioxolane fragment the mesylate, or in some published routes the tosylate forming the diaryl ether bond that completes itraconazole.
That single displacement is the convergent step of the whole synthesis, and it is the reason the two fragments are made separately and joined at the end rather than built up in one linear sequence.
The same triazolone fragment appears in saperconazole, the difluorophenyl analogue of itraconazole, and closely related triazolones with different N-2 alkyl groups appear across the wider Janssen azole patent family.
Applications at a Glance
| Application | Role | Notes |
|---|---|---|
| Itraconazole manufacture | The nucleophilic half; supplies the piperazinyl-phenyl and sec-butyl triazolone portion intact | The dominant commercial application; itraconazole is a large global systemic antifungal generic |
| Saperconazole and related azoles | The same triazolone fragment | Smaller volumes; the dioxolane half differs |
| Triazolone and arylpiperazine building block chemistry | Functionalised phenol with a piperazine linker | Research and development-scale use |
Industry Segments Served
Itraconazole and azole antifungal API manufacturers.
CDMOs and contract synthesis organisations executing client-owned routes.
Analytical and QC laboratories supporting generic filings.

Handling & Storage Guidance
A stable crystalline solid with no significant reactivity hazard. The handling points protect the phenol.
Store cool and dry in tightly closed containers, protected from light and air free phenols oxidise and the oxidation products are coloured.
Allow containers to reach room temperature before opening to prevent condensation.
Minimise open-container time and re-seal immediately after dispensing.
Keep away from strong oxidising agents and strong bases.
Quarantine and re-test any consignment that arrives visibly darker than expected before charging it into a validated batch.
Standard chemical-resistant gloves, eye protection and dust control during weighing.
Consult the Safety Data Sheet supplied with the batch before handling.
Regulatory & Documentation Support
This is a starting material, so a DMF or CEP does not apply to it as a standalone item.
What buyers need is a well-characterised material with documentation that fits their own quality system.
Documentation Supplied
Certificate of Analysis- batch-specific.
Material Safety Data Sheet in GHS format, with destination-language versions on request.
Technical Data Package- manufacturing flow, analytical methods, specification rationale, impurity profile.
GMP Starting Material declaration under ICH Q7 principles, where the GMP grade is ordered.
ICH Q3C residual solvent and ICH Q3D elemental impurity statements.
Certificate of Origin, Non-GMO declaration, BSE/TSE-free and allergen statements.

Frequently Asked Questions
My material looks darker than the last batch is it usable?
Check the assay and the impurity figures rather than judging by eye, but treat darkening as a real signal.
This compound carries a free phenol, phenols oxidise slowly in air and light, and the oxidation products are coloured.
They carry into your coupling and then into crude itraconazole, where colour is a specification attribute. Quarantine anything visibly darker than expected and contact us.
Is this the sec-butyl or the isopropyl triazolone?
The sec-butyl 1-methylpropyl compound, CAS 106461-41-0, which is the itraconazole fragment.
Closely related triazolones with different N-2 alkyl groups appear in the same patent family, including the isopropyl analogue, so confirm by CAS number rather than by name shape.
