Product Overview

1-(6-Methylpyridin-3-yl)-2-[4-(methylsulfonyl)phenyl]ethanone, known throughout the industry simply as the etoricoxib ketosulphone, is supplied under CAS 221615-75-4 with the molecular formula C₁₅H₁₅NO₃S and a molecular weight of 289.35.

It is a deceptively simple molecule: a methyl ketone bridging two aromatic rings, with a 6-methylpyridine on one side and a 4-methylsulfonylphenyl group on the other.

Both of those rings appear unchanged in etoricoxib, and the single methylene between them becomes part of the central pyridine ring of the finished drug.

The compound was first disclosed as a novel intermediate in the late 1990s and has been the subject of considerable process development since, because it sits at a central step of the etoricoxib route.

Grades and specifications are available on request. Every consignment carries a batch Certificate of Analysis and a GHS-format Safety Data Sheet.

Actiza Pharmaceutical Pvt. Ltd. has over 12 years of B2B pharmaceutical export experience, 500+ global clients and a supply footprint across 40+ countries, backed by WHO-GMP and ISO-certified group manufacturing infrastructure in Gujarat, India.

Application in Synthesis

The Final Step

The ketosulphone is cyclised with a three-carbon reagent to close the second pyridine ring and give etoricoxib, 5-chloro-3-(4-methanesulfonylphenyl)-6′-methyl-2,3′-bipyridine.

The methylene carbon of the ketosulphone and its two flanking positions become part of the new ring.

Everything else in the molecule is already in place at this stage.

That is why buying at this point removes so much: the methylsulfonyl group is already installed and at the correct oxidation state, the pyridine methyl is already positioned, and the carbon skeleton is complete.

Handling & Storage Guidance

This is a stable crystalline organic solid with no significant reactivity hazard. The guidance below is about preserving specification.

Store cool and dry in tightly closed containers, protected from moisture and light.

Allow containers to reach room temperature before opening to prevent condensation.

Minimise open-container time and re-seal immediately after dispensing.

Note that the methylene between the carbonyl and the aryl ring is activated, so the compound has an accessible enol form keep it away from strong base during storage.

Keep away from strong oxidising agents and strong bases.

Standard chemical-resistant gloves, eye protection and dust control during weighing.

Consult the Safety Data Sheet supplied with the batch before handling.

Regulatory & Documentation Support

This is a starting material rather than a moiety with its own monograph, so a DMF or CEP does not apply to it as a standalone item.

Because it sits at the penultimate stage, however, buyers will scrutinise it more closely than an early intermediate whatever arrives here has fewer steps in which to be purged.

Documentation Supplied

Certificate of Analysis- batch-specific.

Material Safety Data Sheet in GHS format, with destination-language versions on request.

Technical Data Package- manufacturing flow, analytical methods, specification rationale, impurity profile.

GMP Starting Material declaration under ICH Q7 principles, where the GMP grade is ordered.

ICH Q3D elemental impurity statement.

ICH Q3C residual solvent statement.

Certificate of Origin, Non-GMO declaration, BSE/TSE-free and allergen statements.

Site audit access, subject to prior scheduling and confidentiality agreement.

Frequently Asked Questions

Is this ethanone or ethenone?

Ethanone a ketone. The name is sometimes seen written as “ethenone”, but ethenone means ketene, a completely different and highly reactive species.

The correct name for CAS 221615-75-4 is 1-(6-methylpyridin-3-yl)-2-[4-(methylsulfonyl)phenyl]ethanone. Always confirm by CAS number.

How close to the API is this?

One step. Condensing the ketosulphone with a suitable three-carbon reagent closes the second pyridine ring and gives etoricoxib directly.

The methylsulfonyl group, the pyridine methyl and the full carbon skeleton are already in place.

What is the Minimum Order Quantity?

Our minimum order quantity is available on request.

Qualification quantities for a penultimate-stage intermediate are typically larger than for an early building block, and are discussed at enquiry stage.
